Genetic Testing Center
The Clinical Genetic Testing Center was established in January 2017 on the basis of the Clinical Gene Amplification Laboratory of the Department of Laboratory Medicine, in collaboration with the Department of Pathology. It is located in the Laboratory Center on the 5th floor of Building No. 3 of Zhejiang Provincial Tongde Hospital (total area: 500 square meters). The Genetic Testing Center has 10 professional and technical personnel specializing in clinical laboratory medicine, pathology, molecular biology, immunology, and other fields, including 2 with senior professional titles, 3 with associate senior professional titles, 2 doctorates, and 4 master's degrees. It has established testing technology platforms for molecular biology testing, molecular pathology, gene sequencing, mass spectrometry analysis, and more. The total value of its instruments and equipment exceeds 1000万元, including: 1)PCR-direct sequencing: conventional PCR instruments, an ABI3130 sequencer, and a nucleic acid quantifier. 2)Real-time fluorescence PCR: a fluorescence quantitative PCR instrument, centrifuges, an ultra-clean workbench, thermostatic instruments, etc.; it can perform fluorescence quantitative PCR, ARMS-PCR, HRM-PCR, and more. 3)PCR-gene chips: conventional PCR instruments, hybridization instruments, and a chip reader. 4)In situ PCR or in situ hybridization: an in situ PCR instrument, a fluorescence microscope, thermostatic instruments, etc. 5)Large-scale instruments and equipment for mass spectrometry analysis include AB SCIEX LC-MS 4500, AB SCIEX TripleTOF 4600, and more.
The Center currently carries out more than 40 projects and processes more than 2万 specimens annually.
The main projects include genetic testing and analysis of pathogenic microorganisms (viruses, bacteria, etc., including HBV-DNA quantification, HPV genotyping, HBV P-region gene sequencing, herpesviruses, EV71, gonococcus, Chlamydia trachomatis, MRSA, KPC, influenza A and B viruses, dengue virus, and others); genetic testing of drug targets and drug-metabolizing enzymes (including folate metabolism, clopidogrel metabolism, warfarin metabolism, statin metabolism, allopurinol, adverse drug reactions to carbamazepine, and others); and methylation gene testing related to lung cancer and intestinal cancer. In molecular pathology, it performs HPV testing of cervical exfoliated cells, Her-2 gene amplification testing for breast cancer and gastric cancer, EGFR gene mutation testing for lung cancer, and KRAS, Braf, and Nras gene mutation testing for colorectal cancer, among others.
To meet clinical needs, in November 2018, the Precision Medicine Diagnosis (Research) Center was further established in cooperation with Dian Diagnostics Technology Co., Ltd. The Center gradually began offering second-generation gene sequencing and mass spectrometry analysis projects for clinical use. The mass spectrometry testing projects include psychotropic drug concentration testing, vitamin ADE, B vitamins, bile acid profiles, and others. In genetic testing, on the basis of the original services, it has also launched genetic testing related to psychotropic drug metabolism and added more than 40 tumor genetic testing projects. These projects cover targeted tumor therapy, chemotherapy medications, medication guidance for immunotherapy, and differential diagnosis. They include 9-808-gene panel combinations and whole-exome testing for lung cancer, colorectal cancer, breast cancer, gastric cancer, and lymphoma, such as tumor mutational burden, MSI, PDL-1, and so on.
